Research ProjectDevelopment and fabrication of enzymatic biosensors
Electrochemical biosensors dependent on electron transfer from redox enzymes have been shown to provide exceptional selectivity. Yet, the fabrication of these biosensors is quite challenging. Difficulties in enzyme immobilization and slow electron transfer rate between the enzyme and the electrode have resulted in a limited pool of enzymes used for biosensor fabrication. Successfully fabricated biosensors find a range of applications in various fields such as for clinical diagnosis and food analysis. The aim of my project is to optimize and improve upon the design of enzymatic biosensors. This consists of exploring the use of new enzymes for immobilization onto the microelectrode such as by studying human cytochrome P450 3A4. This implies performing electrochemical measurements on P450 3A4 to quantify its rate of production of hydrogen peroxide. Other aspects of my project include optimizing the deposition of a permselective polymer layer on the electrode, successful and consistent immobilization of the enzyme onto the electrode and finally optimizing the cell patterning strategy for SECM measurements in vitro. |
