Research ProjectDevelopment and characterisation of an amperometric biosensor for ᴅ-serine
R. gracilis ᴅ-amino acid oxidase (RgDAAO) is an efficient enzyme for the oxidation of ᴅ-serine, which generates a stoichiometric amount of hydrogen peroxide during its reaction. Electrochemical analysis of the rate of hydrogen peroxide formation can therefore indicate the activity of the enzyme and the concentration of ᴅ-serine in solution. Fixation of the enzyme onto the tip of a platinum microelectrode allows the efficient probing of ᴅ-serine, with the added special and temporal resolution of a microelectrode. This works looks into improving the response time and the selectivity of the biosensor through the modification of the biosensor manufacture techniques and the thickness of the size exclusion polymer layer. Additionally, the use of scanning electrochemical microscopy (SECM) in conjunction with the enzymatic biosensor allows for the probing of the localized release of ᴅ-serine from a model substrate. Corrosion resistant coatings on steel Hydraulic turbines are essential components in the function of hydroelectric plants. Their use in aqueous environments means that they are subject to corrosion. Self-lubricating polymer materials offer low friction coefficients, but at the cost of low load capacities. In collaboration with Hydro-Québec, this project looks into the assessment of novel composite materials for their suitability, using both experimental methods and simulation of the corrosion processes. Finite element simulation of electrogenerated chemiluminescence (ECL) reactions ECL reactions use an electrochemical reaction to generate a species in an excited state, which then results in the measurable emission of a photon of a specific wavelength. These reactions are multi-stepped, featuring a number of different species, often with short life spans. In order to gain a better understanding of the nature of these reactions, finite element simulations have been run of an ECL reaction which is currently being experimentally studied within our group, with the focus being on the predicted location of reactive species with respect to the electrode surface. |
